at the Nencki Institute of Experimental Biology, Polish Academy of Sciences in Warsaw,
is seeking a highly motivated postdoctoral candidate to work at the interface of stem cell biology, epigenomics, and bioinformatics.
The successful applicant should hold a Ph.D. and have received formal training in molecular biology, stem cell biology, or a related field. Ambition, productivity, and creativity are a must. Preferred candidates should be fluent in English.

Cell differentiation is a stepwise process whereby intrinsic and extrinsic factors modulate gene expression patterns which brings about a cell of a particular identity. Transcriptional regulation is thus at the heart of this process. Distal DNA cis regulatory elements including enhancers impact the activity of the gene promoters thereby regulating the production of mRNA molecules. One of the essential questions is how enhancers find their target genes with precision. CTCF-bound insulators can shield promoters from impact of distal enhancers. Dysfunctions of insulators can lead to diseases including developmental syndromes and cancer. Previously, we have shown that differentiation of pluripotent stem cells is accompanied by a widespread gain of strength of CTCF-bound insulators (Pękowska et a., Cell Systems 2018). The mechanism underlying this phenomenon is unknown.
Scope of tasks:
Our preliminary data revealed a group of protein partners of CTCF that might orchestrate the overall gain of insulatory functions of CTCF in differentiation. We have obtained knockout lines for these factors. Your role will be to address the impact of these proteins on chromatin structure and activity by molecular biology (ATAC-seq, Hi-C) and cell biology approaches (cell differentiation, immunofluorescence, CRISPR-Cas9 mediated construction of reporter cell lines).
