Requirements:

• Doctoral degree in the field of biochemistry, biology or biophysics, obtained not earlier than 7 years before the year of employment in the project (not including maternity leaves).

            • Experience in laboratory work.

• Very good practical knowledge of methods in the field of molecular biology, cell culturing and imaging.

• An important criterion in the selection will also be experience in primary cell culture/ or work with animals, imaging methods (FRAP, FLIM), molecular biology methods, transfection and staining methods of neurons.

            • Scientific achievements documented by publications from the JCR list.

            • Fluent English in writing and speaking.

            • Team work skills.

• High motivation for experimental work. It is expected that during the period of employment at the Institute, Post-doc will not undertake any other work.

 

Scope of tasks :

The aim of our project is to check if and how S-PALM and S-NO profile changes in the brain of mice, which developed depression like symptoms. First we will induce depressive like behavior in mice by exposing them to chronic stress. After achieving behavioral effect animals will receive ketamine at different concentrations in order to achieve antidepressive effect. Finally we will perform sophisticated analysis of the structural changes of dendritic spines in brain structures associated with depression. Neurons will be stained with the lipophilic dye. We will also check the interplay between two modifications and their impact on protein localization in vitro. To investigate the functional role of each investigated molecule modification on dendritic spine morphology we will use primary hippocampal neuronal cultures transfected with plasmid encoding RFP protein. Living dendrites will be imaged before and after treatment with inhibitors using confocal microscopy.

We believe that above-mentioned PTMs, both in qualitative and quantitative manner, play critical role in the pathogenesis of depression as well as other psychiatric disorders. Results obtained by us will help in understanding the function of posttranslational modifications in the nervous system, what may bring us closer to development of more efficient antidepressive therapies.
